基因转染内皮细胞衬里人工血管的研究进展

摘    要:目的:讨论使用外源性抗凝基因修饰内皮细胞后衬里人工血管,使人工血管局部抗凝活性提高以及对内皮细胞其他生物活性的影响等。方法:复习近几年的相关文献,作综述报道。结果:内皮细胞在转染纤溶酶原激活物(t-PA、u-PA、Urokinase)、血栓调节蛋白(TM)和水蛭素(hirudin)等基因后,均使局部血管的抗凝活性增高,但同时也导致内皮细胞粘附性、增殖功能下降。结论:外源性基因能使内皮细胞预衬的人工血管抗凝活性提高,但仍需进行深入研究。

Abstract:Objective To discuss the endothelial cell which was modified by exogenous anticoagulant genes contribute to the increase of antithrombosis activity of lined vascular prosthesis and the influence to other physiological functions of endothelial cells. Methods This summarized paper was made on literature review of recent years.Results The transfection of genes, including plasminogen activator (t-PA, u-PA, Urokinase), thrombomoduline (TM) and hirudin, etc, to endothelial cells resulted in not only the increase of antithrombosis activity of local vascular, but also the decrease of endothelial cell function in adherence and proliferation.Conclusion The increase of antithrombosis activity of lined vascular prosthesis has been done by exogenous genes. However, this technique ought to be studied, intensively.
