摘    要:本文取材于引产胎儿,利用组织块贴壁和胰蛋白酶消化相结合的方法,建立了人肺动脉内皮细胞和成纤维细胞的体外培养;透射电镜观察到内皮细胞含有Weibel-palade(W-P)小体,成纤维细胞内有丰富的粗面内质网和发达的高尔基氏体,但无肌丝和胞间连接。这两种人肺动脉细胞的培养成功,为进一步开展人肺动脉高压机制的研究建立了技术条件。

Abstract:A method using small pieces cultivation in vitro and trypsinization was developed for obtaining endothelial cells and fibroblass from human fetal pulmonary artery. Transmission electron microscopic examination revealed the pressnce of typical WeibelPalade bodies in the cytoplasm and intercellular junctions between the endothelial cells; and the presence of abundant rough endoplasmic reticulum and Golgi complexes as well as the absence of cytoplasmic myofilaments and intercellular junction between the fibroblasts. The cultured human pulmonary artery endothelial cells and fibroblasts may serve as a very useful model for a variety of in vitro studies on the pathogenesis of human pulmonary hypertension.
