大黄素对人血管平滑肌细胞周期蛋白D1表达的影响

摘    要:目的 观察大黄素对人血管平滑肌细胞周期时相和细胞周期蛋白D1 (CyclinD1 )表达的影响 ,探讨大黄素在药物置入物 (涂层支架 )上应用的可能性。方法 取对数增长期的平滑肌细胞 ,采用四甲基偶氮唑盐 (MTT)法观察大黄素对平滑肌细胞增殖抑制的有效浓度范围 ,求出IC50 并干预细胞。然后分别用流式细胞仪和Western blot法进行细胞周期时相和CyclinD1表达的测定。结果 与对照组比较 ,IC50 时药物组G0 /G1 期细胞百分比升高 ,S期细胞百分比下降 ,CyclinD1表达高峰延迟、表达量下调 ,细胞周期受阻于G0 /G1 期。结论 大黄素可以通过下调CyclinD1表达 ,阻滞细胞周期的进程 ,是一种有效的抗平滑肌细胞增殖的药物。

Abstract:Objective To explore the effect of emodin on cyclin D1 and cell cycle phase of human vascular smooth muscle cells cultured in vitro and study the possibility of application of emodin on the drug-eluting stent. Methods The inhibitory effect of emodin on VSMC proliferation was studied by using MTT colorimeter, the effective concentration range and IC 50were calculated, then the cell cycles of the two groups were analyzed by using flow cytometer and cyclin D1 expressions were examined by the techniques of Western-blot. Results Compared with the control group, the percentage of the cells of the drug group in G 0/G 1 phase increased, while that in S phase decreased; the protein expression summit of cyclin D1 was delayed, the dose of protein was decreased, and the cells transition from G 0/G 1 to S was blocked by emodin. Conclusion The inhibition of expression of cyclin D1 may be one of the mechanisms of emodin which is an effective drug for the inhibition of VSMC proliferation.
