肝舒胶囊含药血清体外抗肝纤维化的实验研究

目的研究肝舒胶囊含药血清对肝星状细胞(HSC-T6)自分泌的TGF-β1和Ⅰ型胶原的增生抑制作用,探讨肝舒胶囊体外抗肝纤维化的机制。方法含药血清作用于HSC-T6后,采用貂肺上皮细胞(Mv-1-Lu)生长抑制MTT法检测HSC-T6分泌的TGF-β1的活性及其总量,ELISA法检测PDGF诱导的HSC-T6的I型胶原含量。结果肝舒胶囊含药血清不仅可减少HSC TGF-β1的分泌总量,并抑制TGF-β1的活化,而且还可降低I型胶原的分泌。结论肝舒胶囊含药血清能抑制肝星状细胞TGF-β1产生并活化及降低I型胶原含量,可能是与其具有抗肝纤维化的作用有关。

Experimental study of blood serum containing Ganshujiaonang compound against hepatic fibrosis in vitro

Objective It is to investigate the inhibiting effect of blood serum containing Ganshujiaonang compound on the secretion of TGF-β1 and type I collagen hyperplasia in hepatic stellate cells(HSC-T6),and further investigate its anti-fibrosis mechanism invitro.Methods The activity and total content of TGF-β1 secreted from HSC-T6 fed by the serum contain drugs was measured by mink lung epithelial cell(Mv-1-Lu) proliferation inhibition MTT assay.And type I collagen of the cells induced by PDGF was measured by ELISA.Results Blood serum containing Ganshujiaonang compound could not only decrease the total content of TGF-β1 secreted by HSC-T6 in the supernatant,inhibited its activation,but also could decrease the production of type I collagen.Conclusion Blood serum containing Ganshujiaonang compound can inhibit the production of TGF-β1 in HSC and its activation,decrease the content of type I collage,and which mechanism may be associated with its antifibrotic action.
