大黄素对HSC-T6细胞增殖及细胞周期影响的研究

摘    要:目的 研究大黄素干预对大鼠肝星状细胞株(HSC-T6)增殖和细胞周期的影响,探讨其抗肝纤维化的具体机制。方法 应用大鼠肝星状细胞株伪传代培养,采用不同浓度大黄素进行干预,观察各组细胞在形态学、增殖程度、细胞周期、增殖细胞核抗原(PCNA)表达等多方面的变化。结果 ①5-15Mg/L浓度大黄素干预24h后,对T6细胞的增殖产生显著的抑制效应,同时免疫组化显示PCNA标记指数随着药物浓度的增高其表达阳性率反而逐渐降低。②大黄素同时能够对T6细胞的细胞周期产生阻滞作用,随着浓度的增高,C0/G1期细胞比例逐渐上升。结论 大黄素在体外对于HSC-T6的增殖具有明显的抑制作用,而这种作用可能是通过改变了T6细胞正常的细胞周期来实现的。

Abstract:Objective To investigate the influence of emodin on proliferation and cell cycle of hepatic stellate cell line-T6 and explore its mechanisms of antifibrosis. Methods The rat hepatic stellate cell line, HSC-T6 were generationally cultured in DMEM medium. Entodin of various concentrations were added to the medium in 48 hours.The changes of cell morphol ogy , cell cycle, and expression of PCNA were observed. The effects of emodin on proliferation of T6 cells were observed. Results (1)Emodin markedly inhibited the proliferation of HSC-T6 after 24 hours culture with the concentration of 5-l 5mg/L. The positive rate of PCNA decreas ed with the increase of emodin concentration.(2) The test data from flow cytometer showed emodin can block the cell cycle and along with the increase of concentra tion, the percentage of cells in S and G2/M phase were gradually decreased and that in G 0/G 1 increased. Conclusions Emodin can prevent HSC-T6 cell from proliferation significantly in Vitro probably by changing the normal cell cycle of T6 cells.
