| 摘 要： | 目的研究miRNA在胃癌石蜡包埋组织和新鲜组织的表达。方法应用实时荧光定量PCR检测23例石蜡包埋胃癌组织及与其配对的新鲜冷冻胃癌组织的miR-30c表达,以U-6为内参对照。结果胃癌石蜡包埋组织中的miRNA可以成功扩增出来,胃癌石蜡包埋组织与胃癌新鲜组织中miR-30c表达呈平行性表达(配对t-检验P=0.81)。miR-30c在石蜡包埋组织中的表达量与临床病理因素的关联似乎更紧密。结论可以利用便利获得的存档石蜡组织进行miRNA相关指标表达研究。

| Abstract: | Objective To study the miRNA qualities expression of gastric cancer in the samples formalin-fixed paraffin-embedded （FFPE） and fresh snap frozen. Methods Twenty three cases of gastric cancer embeddied with paraffine, and 23 matched cases non-fixed fresh snap frozened samples were qualitvely miR-30c expressed by ABI 7300 PCR system, U6 was used as the reference gene. Results miRNA extracted from FFPE blocks was successfully amplified using TaqMan^R RT-PCR.Tissue storaged in FFPE appeared to not affect the exprssion number of detected miRNAs compared to matched frozen samples （paired t-test P=0.81）. Correlations between miRNA expression and clinical factors were high across miRNAs in FFPE samples. Conclusion It may be used to take the most readily available archival FFPE cancer samples for the miRNA qualitive expression study. |
