骨髓间质干细胞体外定向肌样分化的研究

摘    要:目的 研究兔骨髓间质干细胞经5—氮杂胞苷诱导,在体外定向肌样分化的情况。方法 取兔胫骨骨髓,分离并培养骨髓间质干细胞,用5—氮杂胞苷苦(10μmol/L)定向诱导,分别在培养的第7、14、2l、28天用免疫组织化学的方法检测细胞中的肌球蛋白重链,取未经5—氮杂胞苷诱导正常培养的细胞为对照组。结果 兔骨髓间质干细胞经5—氮杂胞苷诱导,在其培养的第21、28天免疫组织化学染色方法检测细胞中的肌球蛋白重链呈阳性,而在其培养的第7、14天以及对照组,免疫组织化学染色方法检测细胞中的肌球蛋白重链呈阴性。结论 骨髓间质干细胞是骨髓来源的具有多向分化潜能的干细胞,在5—氮杂胞苷的定向诱导下,可以肌样分化。

Abstract:Objective To study myogenic differentiation of mesenchymal stem cells (MSCs) from rabbbits in vitro after induced by 5-azacytidine.Methods Isolated MSCs from rabbit's bone marrow were cultured in cell cuture medium (control) and in medium sumpplemented with 5-azacytidine (5-aza 10 umol/L).Then MSCs were immunohistologically processed for the expression of myosin heavy chain on the 7th,14th,21st,28th day respectively.Results Only on the 21st,28th day MSCs induced by 5-azacytidine could be identified by the positive staining for myosin heavy chain.On the 7th and 14th day MSCs induced by 5-azacytidine could be identified by the negative staining for myosin heavy chain,and the expression of myosin heavy chain was not detectable on the MSCs in the control group.Conclusion MSCs are one group of stem cells in the bone marrow,with the capacity of differentiating into multiple mesenchymal lineages.They can undergo myogenic differentiation in vitro after induced by 5-azacytidine.
