摘    要:目的:建立RP-HPLC法测定愈肠颗粒中小檗碱和异白茅苷的含量。方法:色谱柱:Diamonsil C18柱(250mm×4.6mm,5μm);测定小檗碱的流动相:乙腈-0.035mol/L磷酸二氢钠(40∶60),测定异白茅苷的流动相:乙腈-0.05mol/L乙酸(用三乙胺调节pH至5.0)(52∶48);检测波长:小檗碱265nm,异白茅苷310nm;流速:1.0 ml/min。结果:小檗碱在1~60μg/ml,异白茅苷在0.1~20μg/ml范围内线性关系良好。小檗碱低、中、高浓度加样回收率分别为(97.24±0.40)%、(97.72±1.39)%、(98.33±2.40)%(n=3);异白茅苷低、中、高浓度加样回收率分别为(103.6±0.01)%、(101.4±0.01)%、(101.8±0.000 7)%(n=3)。3批愈肠颗粒样品中小檗碱的平均含量为(574.13±9.68)μg/g(n=3),异白茅苷的平均含量为(3.09±0.10)μg/g(n=3)。结论:该方法灵敏度高、专属性强,操作简便易行,可用于愈肠颗粒的质量控制。

Determination of berberine and isoimperatorin contents in Yuchang granules by RP-HPLC method

Abstract:Objective: To establish a RP HPI.C method for the determination of berberine and isoimperatorin contents in Yuchang granules. Methods: Chromatographic column: Diamonsil C18 column (250 mm × 4.6 mm, 5 μm); mobile phase for determining berberine: acetonitrile 0.035 mol/L dihydrogen phosphate sodium (40 : 60), mobile phase for determining isoimperatorin : acetonitrile-0.05 mol/L acetic acid (the pH adjusted to 5.0 by triethylamine) ( 52 : 48) ; detection wavelength : berberine 265 nm,isoimperatorin 310 nm; flow rate: 1.0 ml/min. Results: Berberine and isoimperatorin were in good linearity within the ranges of 1 60 μg/ml and 0. 1-20 μg/ml, respectively. Berberine recoveries at low, middle and high concentrations were ( 97. 42 ± 0. 40) %, ( 97. 72±1. 39 ) % and ( 98.33 ± 2.40 ) % (n = 3 ), respectively; and isoimperatorin reseoveries at low, middleandhighconcentrationswere(103.6±0.01)%,(101.4±0.01)%,(101.8±0.000 7)%(n=3),respectively. Average content of berberinewas (574.13±9.68) μg/g (n 3) and isoimperatorin was (3.09±0.10) μg/g(n=3) in 3 batches of Yuchang granules. Conclusion:It is an accurate, sensitive, reproducible method for the determination of berberine and isoimperatorin in Yuchang granules.
