摘    要:目的:观察川崎病(Kawasaki disease, KD)合并冠脉损伤患儿血清对脐静脉内皮细胞(human umbilical vein endothelial cells, HUVECs)增殖及凋亡的影响,以进一步了解KD患儿血管内皮细胞损伤的机制。方法:HUVECs分为正常血清组(Control组)、发热血清组(F组)、KD无冠脉损伤组(Non-CALs组)和KD合并冠脉损伤组(CALs组)。MTT比色法检测细胞增殖,流式细胞术(flow cytometry, FCM)分析各组HUVECs的凋亡情况。结果:MTT比色结果显示,Non-CALs组和CALs组经血清干预后的细胞活性低于F组和Control组,且CALs组低于Non-CALs组,差异有统计学意义(P<0.01)。此外经KD患儿血清作用后,HUVECs的凋亡率增加,与对照组相比差异有统计学意义(P<0.01),且CALs组凋亡率高于Non-CALs组。结论:KD合并冠脉损伤患儿血清具有抑制HUVECs增殖并诱导HUVECs凋亡的作用。

Abstract:Objective To observe the effects on proliferation and apoptosis in human umbilieal vein endothelial cells(HUVECs) cultured with the serum of the children with coronary artery lesions of Kawasaki disease, in order to further understand the possible mechanism of vascular endothelial injury with Kawasaki disease. Methods Human umbilical vein endothelial cells were cultured and were divided into 4 groups: normal serum group, general fever group, Non-CALs group and CALs group. The proliferation was deteeted by MTT test and the apoptosis was deteeted by FCM. Results The OD value of the HUVECs detected by MTT test after being cultured with the serum of the children with Kawasaki disease incluing Non-CALs group and CALs group(0.492±0.033,0.395±0.031)was significantly lower than those of the other two groups (0.592±0.033,0.612±0.036)(P<0.01). The apoptosis of HUVECs detected by FCM after being cultured with the serum of the children with Kawasaki disease was significantly higher than those of the other two groups (P<0.01) and CALs group was higher than Non-CALs group. Conclusions The serum of Kawasaki disease in the acute phase could inhibit the proliferation of HUVECs and induce its apoptosis, indicating that proliferation and apoptosis of endothelial cell play a part in the coronary arter injury of children with KD.
