摘    要:目的]构建真核表达体系,提高抗人乙酰胆碱受体单链抗体(ScFv 637)与人血清白蛋白(HSA)融合基因(ScFv 637-HSA)的蛋白表达质量.方法]扩增ScFv 637-HSA融合基因,经酶切处理后,克隆至真核载体pPIC9 K,线性化处理后,电穿孔入毕赤酵母GS 115.结果]经电泳观察可见相对分子质量为2.6 kbp的融合蛋白ScFv 637-HSA基因和真核表达载体pPIC9 K基因片段.结论]ScFv637-HSA基因已准确地整合到毕赤酵母染色体基因组中.

Abstract:OBJECTIVE To increase the protein expression level of fusion gene of single chain variable fragment(ScFv637) and human serum albumin(HSA) by using eukaryotic expression system.METHODSThe fusion gene(ScFv637-HSA) was amplified and cloned into pPIC9K to construct recombinant vector pPIC9K-ScFv637-HSA.The recombinant vector was linerized and transformed into Pichia Pastoris GS115 by electroporation.RESULTSThe sizes of the fusion gene and the pPIC9K were found to be approximately 2.6kbp and respectively.CONCLUSIONThe eukaryotic expression vector,which bears the fusion gene of ScFv637-HSA,has been successfully constructed.
