吲哚-3-甲醇诱导人鼻咽癌细胞凋亡作用及机制的初步研究

摘    要:背景与目的: 了解吲哚-3-甲醇在体外对人鼻咽癌CNE细胞增殖的影响及其诱导CNE细胞凋亡的作用及其可能机制。 材料与方法: 采用WST-1法检测吲哚-3-甲醇在体外对人鼻咽癌CNE细胞增殖的抑制作用;用Hochest33258荧光染色法及TUNEL法观察吲哚-3-甲醇诱导CNE细胞的凋亡;用Western blotting方法检测凋亡相关信号分caspase-9、caspase-3蛋白表达情况,实验同时设空白对照组及溶剂对照组。 结果: 吲哚-3-甲醇在体外可以明显抑制人鼻咽癌细胞增殖,诱导细胞凋亡并呈剂量依赖性,且在细胞凋亡过程中,半胱天冬酶家族成员caspase-9和caspase-3 活化片段表达阳性。 结论: 吲哚-3-甲醇在体外可抑制人鼻咽癌CNE细胞增殖并诱导其发生凋亡,且凋亡的发生可能与caspase-9、caspase-3活化有关。
