TNP-470对小鼠腹水瘤腹水生成的影响

目的]研究血管生成抑制剂TNP-470对小鼠腹水瘤细胞腹腔种植的影响。方法]80只615小鼠腹腔内接种腹水瘤细胞株后，随机分为5组，然后分别腹腔内注射生理盐水（第1组），DDP（第2组）及不同剂量的，TNP-470（第3、4、5组），观察瘤细胞株对各组小鼠的腹水生成，瘤细胞数及生存时间；透射电镜观察TNP-470作用后腹腔内肿瘤细胞的形态学变化。结果]与生理盐水对照组相比，DDP、不同剂量的TNP-470可显著抑制615小鼠腹水的生成，减少瘤细胞数量，延长生存期，DDP组、TNP-470低剂量组、TNP-470中剂量组、TNP-470高剂量组生存时间分别为26．25、19．75、28．38、21．25d，与对照组生存时间14．88d相比，有显著性差异（P〈0．05），且中剂量TNP-470可使小鼠寿命明显延长，平均可达28．38d。结论]TNP-470能抑制小鼠腹水瘤腹腔种植及腹水生成，不同程度的延长生存期。

Study of inhibitory effect of TNP-470 to ascitogenesis of ascitogenous tumor cells in mice

Objective]The aim of this study to observe the effect of TNP-470 on mice implanted intraperitoneally with ascitogenous tumor cells and investigate its mechanism.Methods] Eighty 615 mice were injected with ascitogenous tumor cell line and were divided into 5 groups at random: Group 1,normal saline;Group 2,Cisplatin;Group 3~5,various concentration of TNP-470.Production of ascites,number of tumor cells and survival period were evaluated.Morphological changes of tumor cell were observed with eletromicroscope.Results]Cisplatin and various concertration of TNP-470 obviously inhibited the production of ascites and prolonged the survival period of tumor-loading mice.Survival period of cisplatin,low dose TNP-470,middle dose TNP-470 and high dose TNP-470 was 26.25,19.75,28.38,21.25 d respectively.Comparison of above groups with group 1 showed significantly difference(P<0.05).The middle dose of TNP-470 obviously prolonged the survival period of tumor-loading mice,the survival period was 28.38 d.Conclusion]TNP-470 could inhibit or eradicate mice following intraperitoneal implatation of ascitogenous tumor cells and production of ascites and prolong the survival period of mice that suffered from malignant ascites.
