骨髓基质细胞经不同移植途径在损伤脊髓中的迁移

摘    要:目的 观察骨髓基质细胞(BMSCs)经不同移植途径在损伤脊髓中的迁移和分布规律.方法 40只Wistar大鼠随机分成4组,以改良Allen法制备脊髓损伤模型.第3代BMSCs经CM-Dil荧光染料标记后按不同途径移植.A组:脊髓损伤后行第四脑室注射移植;B组:脊髓损伤后行损伤区蛛网膜下注射移植;C组:脊髓损伤后行损伤区远端蛛网膜下注射移植;D组:脊髓损伤后行股静脉注射移植.分别于移植后24 h、1、2、3、4周取损伤脊髓段作冰冻切片,倒置荧光显微镜下观测移植的BMSCs在损伤脊髓中心的聚集情况.结果 移植后24 h:除B组外脊髓损伤区未发现红色荧光;移植后1周,A、C组移植的BMSCs开始向脊髓损伤区迁移;移植后2周,除D组外,各组BMSCs迁移、聚集在损伤脊髓的中心;移植后3-4周,脊髓损伤区BMSCs数量减少,B组减少速度更快.结论 经第4脑室、硬脊膜下移植的BMSCs能迁移聚集在损伤脊髓的中心,经静脉移植的BMSCs极少迁移到损伤脊髓的中心.

Migration and distribution of bone marrow stromal cells in injured spinal cord with diffferent transplantation techniques
Abstract:Objective To investigate migration and distribution of bone marrow stromal cells in injured spinal cord with intradural space transplantation. Methods Forty Wistar rats were randomly as-signed to 4 groups. The spinal cord injury model was prepared according to the modified Allen method.Bone marrow stromal ceils were labeled by CM-Dil. 5.0 × 106 cells were transplanted with different tech-niques including intraventricular injection (group A), injured spinal cord intrathecal injection (group B),remote intrathecal injection (group C) , and intravascular injection (group D). Spinal cord was dissected at 24th h, 1st,2nd,3rd and 4th week after transplantation. Four-μm thick sections were cut on a cryostat and observed under the fluorescence microscopy. Results Twenty-four h after transplantation,no fluorescence was observed in SCI center except group B. One week later,BMSCs in groups A and C began to migrate to the injured parenchyma;2 weeks later, B MSCs penetrated into the injured parenchyma except group D;3-4 weeks later,the number of BMSCs in injured spinal tissues was reduced,and the number of BMSCs in group B was decreased faster than in groups A and B. Conclusion BMSCs transplanted through intravent-ricular injection,injured spinal cord intrathecal injection and remote intrathecal injection could migrate to the injured parenchyma of spinal cord effectivelly.
