摘    要:目的:观察GM-CSF基因转导的肿瘤细胞免疫小鼠的抗肿瘤效果。方法:用电穿孔法将小鼠GM-CSF的表达质粒导入RMA淋巴瘤细胞,经G418筛选后,将GM-CSF高表达克隆(RMA-GM)细胞皮下接种C57BL/6小鼠,观察成瘤性,以及经丝裂霉素-C处理灭活的RMA-GM细胞免疫小鼠,观察抗肿瘤作用。结果:RMA-GM细胞免疫小鼠及亲代肿瘤细胞攻击后与对照组相比,肿瘤生长速度减慢,小鼠生存期明显延长

Abstract:Objective:To evaluate the antitumor effect of GM CSF gene transfered Tumor Vaccine Methods:A mouse GM CSF expressing plasmid was transferred into RMA lymphoma cell line by electroporation After screening by G 418 ,the high titer GM CSF expressing cell clone (RMA GM) were injected into the C56BL/6 mice subcutaneously Tumor formation was evaluated Mice were immmunized with mitomycin C inactivated RMA GM cells and antitumor effects were evaluated Results:Tumor formation in C57BL/6 mice inoculated by the RMA GM cells was delayed slightly After vaccination with mitomycin C inactivated RMA GM cells,the mice were challenged by parental tumor cells Tumor development speed was reduced;survival time was remarkably delayed compared with the control groups and 40% mice without tumor were in long term survival Conclusion:GM CSF gene transfered tumor vaccine in RMA tumor model induced better antitumor immune effect and will be a novel approach to cancer in the future
