摘    要:目的:观察中药“脊髓康”对大鼠脊髓急性损伤后脊髓组织Nogo-A表达的影响。方法:选用162只SD大鼠,随机分成假手术组、模型组、强的松组、脊髓康高剂量组、脊髓康中剂量组、脊髓康低剂量组,每组27只,分别于干预后3天、7天、14天处死大鼠,以免疫组化、Western Blot及荧光定量PCR检测大鼠脊髓组织中Nogo-A的表达量。结果:脊髓损伤各组在损伤后3天时Nogo-A蛋白和mRNA表达较低,7天后迅速上升达到高峰,至14天逐渐下降。与模型组比较,脊髓康中剂量组和强的松组在损伤后3天、7天、14天时,Nogo-A蛋白和mRNA表达差异均有统计学意义(P<0.05)。结论:中药“脊髓康”能有效抑制大鼠脊髓损伤区Nogo-A表达,有利于脊髓损伤修复。

Abstract:This study was aimed to observe the effect of Chinese medicine Ji-Sui-Kang (JSK) on the expression of Nogo-A in acute spinal cord injury rats. A total of 162 SD rats were randomly divided into the sham-operated group, model group, prednisone group, JSK group of high, middle and low dose, with 27 rats in each group. The rats were sacrificed on the 3rd, 7th and 14th day after intervention, respectively. The expression of Nogo-A of spinal cord tissues among all rats was detected by immunohistochemistry, western blot and fluorescence quantitative PCR. The results showed that Nogo-A protein and mRNA expression of spinal cord injury rats were decreased on the 3rd day after injury. However, it increased rapidly to the climax on the 7th day. And then, it gradually decreased on the 14th day. Compared with the model group, there were statistical differences on Nogo-A protein and mRNA expression of rats in the middle dose JSK group and prednisone group on the 3rd, 7th and 14th day after injury (P<0.05). It was concluded that Chinese medicine JSK can effectively inhibit the expression of Nogo-A in acute spinal cord injury rats. It promoted the nerve regeneration in the spinal cord.
