摘    要:目的:探讨冲击波对体外培养的鼠成骨细胞的增殖及分化能力的影响。方法:将原代培养的成骨细胞随机分成冲击波组和对照组,用不同能量(5、10、15、20 kV)冲击波处理后接种于96孔培养板,根据细胞存活率和增殖情况确定最佳的冲击波能量值。将传4代成骨细胞用冲击波处理后,用CCK-8法、细胞分泌碱性磷酸酶试剂盒、茜素红染色对两组细胞增殖及分化进行测定。结果:冲击波处理体外原代培养成骨细胞的最佳能量值为10 kV(500次),CCK-8法检测显示,冲击波组细胞增殖速度快于对照组(P〈0.001),细胞分泌碱性磷酸酶高于对照组(P〈0.001),茜素红染色显示,冲击波组矿化结节面积明显大于对照组(P〈0.001)。结论:最佳能量冲击波可促进成骨细胞的增殖与分化。

Abstract:Objective: To study the effects of shock wave on proliferation and differentiation of rats'osteoblasts in vitro.Methods:Primary osteoblasts were isolated from calvaria of neonatal rat and cultured in 96-well plate,and an optimal energy of shock wave was selected according to survival and proliferation of osteoblasts treated by different energies(5、10、15、20 kV)of shock wave.The optimal energy of shock wave treating osteoblasts was 10 kV(500 pulse),which had the best promotion effect on the osteoblasts survival and proliferation.After the 4th generation osteoblasts treated by optimal energy of shock wave,the proliferation and differentiation of osteoblasts were dertermined by CCK-8 Kit,AKP kit and Alizarin red staining.Results:The cell growth,alkaline phosphatase activity and cell mineralization in the shock wave group were increased,compared with the control group(P0.001).Conclusion:The optimal shock wave can promote the proliferation and differentiation of osteoblasts.
