牛磺酸和硫酸镁单用及合用抗缺血/再灌心律失常作用

摘    要:本实验采用麻醉大鼠缺血/再灌心律失常模型,研究了牛磺酸(Taur)和硫酸镁(MgSO_4)单用及合用抗再灌心律失常作用,并观察了MgSO_4及MgSO_4与Taur合用对正常大鼠在体血流动力学的影响。结果表明:Taur 50 mg.kg~(-1),MgSO_4 25 mg.kg~1具有部分抗心律失常作用,Taur 100mg.kg~(-1),150 mg.kg~(-1),MgSO_4 50 mg.kg~(-1)、100 mg.kg~(-1)抗心律失常疗效增加,两药小剂量合用作用较各单用药组明显增强。MgSO_4 25mg.kg~(-1)对大鼠心肌仅有轻度负性肌力作用,与Taur 50 mg,kg~(-1)合用,负性肌力未见增加,但具有负性频率和减低心肌耗氧量作用,提示其抗心律失常作用与心脏保护有一定关系。

Abstract:The effect of taurine (Taur) alone and in combination with magnesium sulfate (MgSO4) on ischemia/reperfusion arrhythmia was investigated, The arrhythmias were pruclucecl by coronery artery occlusion for 10 min followed by reperfusion. In addition , the presdent study also observed the effects of MgSO4alpne and in combination with Taur on hemodynamics.The results showed that Taur (50 mg. kg 1)and MgSO4(25mg. kg1 had partly antiarrythmic effects. Taur (100mg. 150mg, Kg 1) MgSO(50mg, 100mg. kg 1) had singniricantly effects of antiar-rhythmia,Taur (50mg,kg 1)combied with MgsO4(25mg. kg1)shorten the during time of ventricular tachycardia(VT)more than either drug did alone. The hypotensive effect of MgSO4(25mg. kg 1)did not be increased by co -adiministration of Taur,but had the effect of reducing oxygenic consumption in myocardium.These finding indicated that Taur in combination with MgSO4 were moer effect on reperfusion an rhythmia. The mechanism of antiarrhvthmic effect of Taur and mgso4 may be involved in the effect of aefence of myocardium.
