干酪素法测定大黄鞣质类成分的研究

摘    要:目的:建立稳定的干酪素法测定大黄鞣质的方法。方法:用干酪素法测定大黄鞣质。优化条件为:以30%甲醇回流提取30min,干酪素用量600mg,1.5%Na2CO3显色后15min测定。结果:优化方法线性关系良好.鞣酸平均回收率为99.27%,RSD=1.00%(n=3)。结论:该法简单、灵敏、稳定,是测定大黄鞣质较好的方法。

Abstract:Objective:To establish a stable method for determining the content of tannins in Rheum officinale by casein method.Methods:The casein method was used to determine the content of tannins.The optimum conditions of determination were using 30% methyl alcohol as extracting solvent, refluxing 30 min, adopting 600 mg caseins, adding 1.5% Na 2CO 3 and determining after 15 min.Results:The selected method had a good linear dependence. The average recovery of tannic acid was 99.27 % with RSD =1.00%( n =3).Conclusion:It is simple, sensitive and reproducible to determine the content of tannins in Rheum officinale .
