失神经肌萎缩微血管床的变化

摘    要:目的观察失神经支配后骨骼肌微血管床的变化,探讨延缓失神经肌萎缩可能的机制。方法采用切断右侧坐骨神经的方法建立大鼠失神经支配骨骼肌萎缩的模型。通过单宁酸一氯化铁媒染,结合HE染色、天狼星红染色,观察大鼠术后4周、6周正常侧与实验侧毛细血管密度、胶原纤维面积与肌纤维横截面积比值、腓肠肌湿重比。结果大鼠腓肠肌湿重比随失神经支配时间延长降低,实验侧毛细血管密度较对照侧显著减低(P〈0.05),而胶原纤维面积与肌纤维面积比值高于对照侧(P〈0.05)。结论随着失神经支配时间的延长,骨骼肌微血管密度减低,胶原纤维增生,影响骨骼肌的血供,直接影响到肌萎缩的发展。

Abstract:Objective To explore the mechanism of delaying denervated muscle atrophy by observing the microcirculatory bed changes of denervated rat gastrocnemius.Methods The sciatic nerve cut off the right method to set up the rat lost control of skeletal muscle atrophy neural model.Through the tannic acid-ferric chloride mordant dyeing,combined with HE dyeing,sirius red dye,the rats after surgery,the 4 weeks,6 weeks after surgery,the normal side and experimental side capillary density,collagen fiber area and muscle fiber cross-sectional area of wet weight ratio,gastrocnemius than were observed.Results Rats with wet weight loss than gastrocnemius muscle neurogenic control extended time was reduced,experimental side capillary density than control side significantly reduced(P < 0.05),but the collagen fiber area and the percent of ratio than the control side(P < 0.05).Conclusion With neurogenic control lost the extension of time,skeletal muscle microessel-density reduced,collagen,the influence of skeletal muscle blood supply,it is directly affects the development of muscle atrophy.
