绞股蓝对D-半乳糖所致亚急性衰老大鼠下丘脑的抗衰老作用及其机制研究

摘    要:目的:观察绞股蓝对衰老大鼠下丘脑的保护作用,并探讨其作用机制。方法:40只SD雄性大鼠,按体重随机分为4组,每组10只:正常对照组、衰老模型组、生理盐水灌胃组、绞股蓝治疗组(100mg.kg-1.d-1)。分别称量大鼠脾脏、胸腺指数,检测超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、一氧化氮合酶(nitric ox-ide synthase,NOS)、一氧化氮(nitric oxide,NO)的活性变化。结果:衰老模型组大鼠下丘脑匀浆中SOD活性明显降低,MDA,NO活性明显增高,同正常对照组比较有显著性差异(P<0.01)。给予绞股蓝后可明显提高SOD活性,同时MDA,NO活性降低明显(P<0.01)。与衰老模型组比较有显著性差异(P<0.01)。结论:绞股蓝能提高抗氧化物酶的活性,有效抑制衰老大鼠下丘脑氧自由基的形成,增加免疫器官重量,并可拮抗下丘脑NO引起的神经毒性,从而延缓D-半乳糖所致的大鼠衰老。

Abstract:Objective: To study the effect machanism of Gynostemma pentaphyllum Makino to hippocampus in aged rats induced by D-galactose.Methods: 40 SD rats were randomly and egually divided into four groups:group 1: Normal,group 2: model,group3:given gastriclavage with Salines,and group4:with Gynostemma pentaphyllum Makino(100mg/kg).After 40 days,the activities of SOD,MDA,NO,NOS were measured with colorimetric method in hippocampus,and the changes of weight and the index of thymus and spleen were observed.Results: In group 2 the content of SOD was lower and the activities of MDA,NO and NOS were higher than group 1(P<0.01).In group 4 the SOD activity was significantly increased,the activities of MDA,NO,NOS were decreased,the weight of rats had no change significantly(P>0.01),and the index of thymus and spleen increased significantly than that in group 1(P<0.01).Conclusion: Gynostemma pentaphyllum has strong free radical that has scavenging activity in hippocampus,and may increase the weight of immunity organs and restrain the activities NO.
