摘    要:目的:探讨高压氧(Hyperbaric oxygenation,HBO)对兔耳增生性瘢痕形成及对血管内皮生长因子(VEGF)、转化生长因子β1(TGFβ1)表达的影响。方法:选取20只新西兰大白兔建立兔耳瘢痕模型,每耳6个创面,随机分成实验组和对照组2组,每组10只,共120个创面,实验组术后立即给予HBO处理,对照组处于常压环境中。术后第29天切取瘢痕组织,测量瘢痕增生指数(hypertrophic index,HI),并用HE染色观察瘢痕形态学变化,免疫组织化学方法检测TGFβ1及VEGF的表达。结果:实验组HI值明显低于对照组(P〈0.01);HE染色结果示实验组成纤维细胞数目较对照组明显减少(P〈0.01);免疫组织化学检测结果示实验组VEGF、TGFβ1的表达量较对照组显著降低(P〈0.01)。结论:HBO可降低VEGF、TGFβ1的表达,对兔耳增生性瘢痕有明显抑制作用。

Abstract:Objective To explore the influence of hyperbaric oxygen on the scar formation at the rabbit ears and the expression of VEGF and TGFβ1. Methods 20 New Zealand rabbits were used to establish the hypertrophic scar model on the ears.The rabbit were randomly divided into experimental group (n=lO) and control group (n=10).The rabbits in the experimental group received hyperbaric oxygen treatment after the operation. After day 28 the scars were collected. Histomorphological change of scars were observed by hematoxylin-eosin staining and the expression of VEGF and TGFβ1 was detected by immunohistochemical method. Results The number of fibroblasts in experimental group were significantly reduced as compared with those in control group. The expression of VEGF and TGFβ1 could be detected with a distinctly down-regulated in experimental group. Conclusion The hyperbaric oxygen can effectively reduce the expression of VEGF and TGFβ1 and inhibit the early hyperplastic scar in rabbit ears.
