雷诺昔芬对骨质疏松大鼠骨髓基质细胞的影响

摘    要:目的:研究不同浓度的雷诺昔芬对骨质疏松大鼠骨髓基质细胞(bone marrow stromal cells,BMSCs)增殖及分化的影响。方法:构建Sprague-Dawley(SD)大鼠骨质疏松模型,培养假手术组(Sham-ovariectomized rats,Sham组)及手术组(ovariectomized rats,OVX组)大鼠BMSCs,取生长良好的第3、第4代BMSCs,加入不同浓度的雷诺昔芬作用后,以MTT法检测BMSCs的增殖,速率法检测碱性磷酸酶(alkaline phosphatase,ALP)的含量。结果:Sham组大鼠第4代BMSCs较第3代增殖缓慢,ALP含量无明显变化;OVX组大鼠第4代BMSCs增殖能力增强,但ALP含量亦无明显变化。结论:OVX组BMSCs的增殖活力、成骨分化能力显著高于Sham组。雷诺昔芬可促进Sham组BMSCs的增殖,可促进OVX组BMSCs的增殖与分化。

Abstract:Objective: To investigate the effects of different concentration of raloxifene on the proliferation and differentiation of bone marrow stromal cells (BMSCs) from the experimental rats. Methods: Osteoporosis models in Sprague-Dawley (SD) rats were established. BMSCs from the Sham-ovariectomized rats(Sham) and the ovariectomized rats(OVX) were cultured in vitro. BMSCs between passages 3,4 were cultured under different concentration of raloxifene. The proliferation of BMSCs was evaluated by MTT and the differentiation by comparing the synthesized alkaline phosphatase(ALP)was tested by rate method. Results: The 4th passages(P4) BMSCs from the Sham had weaker ability of proliferation than the 3rd passages (P3). But the P4 BMSCs from the OVX had increased ability of proliferation. For the differentiation, there was no statistical difference between the P3 and P4 BMSCs from both group rats. Conclusion: The ability of the proliferation of BMSCs from the Sham dropped quickly after subculturing. The BMSCs from the OVX had the much stronger ability of proliferation and synthesized much more ALP. Raloxifene could promote the growth of the P4 BMSCs from both group rats, but just promoted the differentiation of the BMSCs from the OVX.
