miR-218下调与宫颈癌发生相关性的初步研究

目的初步探讨miR-218与宫颈癌发生发展以及浸润转移的相关性。方法选取25例正常宫颈组织、35例宫颈上皮内瘤样病变组织和52例宫颈癌组织,通过实时荧光定量PCR法检测miR-218的相对表达量,初步探讨miR-218的表达与宫颈癌的发生关系。结果与正常宫颈组织相比,宫颈上皮内瘤变组织和宫颈癌组织的miR-218表达量均下降,差异有统计学意义(P<0.05)。与宫颈上皮内瘤变组织相比,宫颈癌组织的miR-218表达量下降,差异有统计学意义(P<0.05)。比较肿瘤直径大小、不同年龄、临床分期和绝经与否的宫颈癌组织中miR-218表达水平差异均无统计学意义(P>0.05);而比较不同的病理分级、间质浸润深度、淋巴结转移与否在宫颈癌组织中miR-218相对表达水平,差异均有统计学意义(P<0.05)。结论 miR-218的表达与宫颈癌的发生发展、转移和侵袭有关,为肿瘤靶向治疗提供了新靶点,并可作为治疗宫颈癌及评价预后的参考指标。

Preliminary research on the correlation between miR-218 down-regulation and cervical cancer

Objective To discuss the correlation between miR-218 and cervical cancer development as well as the invasion of metastasis. Methods Selected 25 cases of normal cervical tissue, 35 cases of cervical intraepithelial neoplasia tissue and 52 cases of cervical cancer tissues,to detect the relative expression level of miR-218 by real-time fluorescence quantitative PCR, then studied the relationship between preliminary study of miR-218 expression and cervical cancer relationship. Results Compared with normal cervical tissue, the expression levels of miR-218 in cervical intraepithelial neoplasia and cervical cancer tissues were decreased, the difference was significant of statistics （P 〈 0.05）. Samely, compared with cervical intraepithelial neoplasia, the expression of cervical cancer miR-218 was decreased, the difference was also significant of statistics （P 〈 0.05）. The miR-218 expression levels of cervical cancer with different pathological grade, stromal invasion depth and-lymph node metastasis had no statistically significant difference （P 〉 0.05）. However, miR-218 expression level of cervical cancer with different systematws of pathology, stromal invasion depth, lymph node metastasis, had significant difference of statistics （P 〈 0.05）. Conclusion The expression of miR-218 is related with the development, metastasis and invasion of cervical cancer, which can provide a new target for cancer therapy, and can as a reference index for medical treatment of cervical cancer and the evaluation of the prognosis.
