高侵袭力人胃癌细胞亚系MKN-28s的建立

目的以穿透人工基底膜的能力大小为依据筛选具有体外高侵袭能力的胃癌细胞亚系。方法用体外培养法收集扩增能侵袭穿透人工基底膜的胃癌细胞亚系;比较母亚两系的侵袭能力;流式细胞术检测母亚两系细胞周期;免疫组织化学SP法观察母亚两系细胞Flt-4蛋白表达情况。结果从母系MKN-28中成功筛选出高侵袭力胃癌细胞亚系MKN-28S;显微镜下细胞计数示亚系侵袭至膜背面的数量较母系明显增多（P〈0.05）,细胞侵袭力增强;亚系中Flt-4蛋白表达同母系相比显著增高;亚系增殖指数（PI）高于母系。结论MKN-28S细胞较MKN-28细胞具有更强体外侵袭力和增殖能力。

Establishing of Gastric Carcinoma Subcell Line(MKN-28S) With Higher Etastatic Potential

Objective Specially constructed in vitro invasion chamber cells that penetrate the matrigel were selected and cultured to establish offspring cell lines. Methods Matrigel invasion assay was used for selection of tumor subline ; the etastatic ability of cells was investigated with matrigel invasion assay;Fit -4 protein in MKN - 28S was detected by immunohistochemical staining;Flow cytometry was used to test cell cycle. Results A gastric carcinoma cell subceU line （ named MKN -28S） was established from its parent cell line MKN - 28 with higher metastatic potential. Compared with parental ceils, the migrative ability of passing through the membrane millipore was obviously increased in MKN -28S （P 〈 0.05 ）. The expression of Fit - 4 increased significantly in the screened subcell line. And the proliferating index （ PI ） of MKN - 28S was higher than that of MKN - 28. Conclusions This research suggested that MKN - 28S cell had more powerful proliferation and stronger etastatic ability than that of its mother line MKN - 28.
