急性冠状动脉综合征患者外周血T细胞Rho激酶活性检测的意义

目的研究急性冠状动脉综合征(ACS)患者外周血T细胞中Rho激酶活化情况及其临床意义。方法选择ACS患者270例,根据冠状动脉造影和临床诊断分为稳定性心绞痛(SAP)组85例、不稳定性心绞痛(UAP)组95例,急性心肌梗死(AMI)组90例和非冠心病患者(对照组)70例。冠状动脉病变程度采用冠状动脉Gensini积分量化。外周血采用RosettSep T细胞提取试剂盒。磷酸化肌球蛋白磷酸酶靶亚基1(MYPT1)蛋白表达来表示Rho激酶活性,磷酸化MYPT1蛋白检测采用免疫印迹法,细胞因子水平采用酶联免疫吸附试验检测。结果 UAP组和AMI组外周血T细胞Rho激酶活性明显高于SAP组和对照组(P<0.05)。Rho激酶活性抑制剂Y27632对ACS患者T细胞白细胞介素6分泌有明显抑制作用(P<0.01),对白细胞介素10分泌无明显抑制作用。Gensini积分≥20分患者外周血T细胞Rho激酶活性明显高于Gensini积分<20分患者(P<0.05)。结论 ACS患者T淋巴细胞存在Rho激酶活化异常,并且与ACS患者冠状动脉斑块的稳定性和病变严重程度有关。

Clinical significance of Rho kinase in peripheral blood T cells from patients with acute coronary syndromes

Objective To investigate the activity of Rho kinase in peripheral blood T cells of patients with acute coronary syndromes and to explore the clinical significance of this activity in the development of atherosclerosis.Methods ACS patients(270 cases) were divided into SAP(85 cases), UAP(95 cases) and AMK90 cases) groups,with other 70 patients having negative result in coronary angiography as the control group.The severity of angiographic CAD was assessed by Gensini score.T cells were isolated by RosettSep T cell purification kit.Rho kinase activity was assessed by Western blot.Cytokine production was examined by ELISA.Results The activity of Rho kinase in ex vivo T cells from patients with UAP and AMI was significantly increased compared with the control group and SAP group(P < 0.05).Secretion of IL-6 in cultured T cells from CAD patients was inhibited by Y27632,a specific inhibitor of Rho kinase,however,IL-10 secretion was not affected.The Rho kinase activity of T cells from CAD patients with high Gensini score was significantly higher than that from patients with low Gensini score.Conclusion The ex vivo T cells from patients with CAD have increased activity of Rho kinase.Alteration of Rho kinase activity is correlated with the severity of CAD and may contribute to the pathogenesis and development of atherosclerosis.
