以外周血T淋巴细胞核仁形成区嗜银蛋白的表达评价肿瘤患者免疫状态

摘    要:目的:比较并评价外周血T淋巴细胞核仁形成区嗜银蛋白(Ag-NORs)的表达以及T淋巴细胞表面分化抗原和自然杀伤细胞(NK)等分别在肿瘤患者免疫状态评价中的应用.方法:对外周血T淋巴细胞经培养后制片进行银染,通过KL型肿瘤免疫图像分析系统计数淋巴细胞核仁银染面积与细胞核银染面积比值(I.S%);应用流式细胞仪检测外周血T淋巴细胞表面分化抗原(CD3、CD4、CD8、CD4/8)及NK细胞.结果:在健康人组和肿瘤患者组之间, 外周血T淋巴细胞Ag-NORs及NK细胞差异有显著性,而CD3、CD4、CD8、CD4/8差异无显著性 ;以恶性肿瘤发病概率分别与外周血T淋巴细胞的Ag-NORs的表达以及CD3、CD4、CD8、CD4/ 8、NK建立回归方程,以两个回归方程判断恶性肿瘤发病的有效性,并画出ROC曲线,A g-NORs的有效性和ROC曲线下面积均高于T淋巴细胞表面分化抗原及NK细胞.结论 :从监测机体免疫状态的角度出发,外周血T淋巴细胞Ag-NORs的检测较以往T淋巴细胞表面分化抗原以及NK细胞的联合检测更为优越.

Abstract:Objective: To compare and evaluate the application in tumor immunity of argyrophilic nuclear organizer region associated proteins (Ag NORs) and differentiation antigen(CD3,CD4,CD8,CD4/8) of peripheral T lymphocytes and nature kill cells(NK). Methods: Silver staining was conducted by one step method, then the odds of lymphocyte nucleolus area and nucleus area were counted by KL tumor immune image analysis system. CD3, CD4, CD8, CD4/8 and NK were detected by flow cytometer. Creation regression equation was used between episode probability and Ag NORs or CD3,CD4,CD8,CD4/8 and NK. Then the effectiveness was calculated and ROC curve drawn. Results: Between tumor patients and healthy subjects, there was significant difference in Ag NORs and NK. The effectiveness and the area under ROC curve of Ag NORs were larger than those of the other. Conclusion: For a marker of monitoring body's immune state in screening malignancy, Ag NORs of T cells are better than combination of CD3, CD4, CD8, CD4/8 and NK.
