摘    要:本研究应用Dot-ELISA方法,以卫氏并殖吸虫(Pw)成虫单克隆抗体A_3D_3(McAbA_3D_3)检测5只实验感染犬吡喹酮治疗前后血清循环抗原(CAg)动态。结果表明,检出CAg最早时间为感染后第4d,至第19d,4只犬CAg检测均为阳性。感染后第101d,其中2只以吡喹酮总量3.2g进行治疗,治疗后短期内CAg滴度明显升高,至治疗后第6d达高峰,1号犬和3号犬分别在治疗后第65d和26dCAg滴度下降为0。2只未治疗感染犬CAg滴度维持于1:160~1:640。治疗后80d剖检2只治疗犬,其肺部均有多个陈旧性虫囊,内多为变性坏死虫卵,未发现虫体。未治疗犬于死后解剖获成虫60条。上述研究结果提示用McAbA_3D_3作Dot-ELISA对犬的Pw活动性感染具有早期诊断和疗效考核价值。

Abstract:The observation on kinetics of circulating antigen (CAg) before and after praziquantel treat- ment in dogs infected with Paragonimus westermani was conducted with McAb A_3D_3 and Dot- ELISA. The earliest time when CAg could be detected was 4d after infection (Dog No.4). CAg of all 4 dogs (No. 1,2,3,4) were positive at 19d after infection. Dog No.1 and 3 were treated with 3.2g praziquantel at 101d after infection. CAg titres were distinctly increased shortly after an effective treatment with peak appearing 6d post-treatment, and it began to decline thereafter and became 0 at 65d and 26d respectively after treatment. No worm was found in treated dogs and sixty alive adult worms were harvested in the cysts of lungs from infected and non-treated dog No.2 at 137d after infection. The results showed that McAb A_3D_3 Dot-ELISA might be applied for early stage diagnosis of paragonimiasis and efficacy evalution of treatment.
