视网膜母细胞瘤的DNA含量研究

目的 :研究不同类型视网膜母细胞瘤 (Retinoblastoma ,Rb)标本中的DNA含量及细胞周期各时相分布 ;探讨影响Rb增殖和分化的因素。方法 :应用流式细胞学技术检测 2 0例Rb标本的DNA含量及细胞周期各时相分布 ,定量分析其增殖指数 ,并将各指标进行比较。结果 :正常视网膜组织细胞与不同分化类型Rb的DNA组方图明显不同。Rb的DNA指数 (DI)及S期细胞百分比 (SPF)在不同的Rb视神经侵犯程度、分化类型中具有显著的统计学差异 :Rb累及视神经筛板后的Rb组的DI和SPF值均高于未累及筛板组 (P <0 0 5 ) ;分化型Rb的DI明显低于未分化型组 (P <0 0 1) ,而SPF明显高于未分化组 (P<0 0 1)。结论 :视网膜母细胞瘤DNA含量的研究有助于认识Rb的发生、发展规律 ,具有一定的理论价值与应用前景

The study on DNA content of retinoblastoma

Objective:To study the DNA content and cell cycle in different types of retinoblastoma(Rb) and analyze the proliferation exponent.To evaluate the factors that influence the proliferation and differentiation of Rb.Methods:In 20 Rb specimens,the DNA content and cell cycle of Rb were also studied with flow cytometry(FCM)and the proliferation exponent of tumor was analyzed.All data were analyzed statistically.Results:Between normal retina tissue and different differentiation of Rb,there were significant differences of DNA diagrams,and also existed between DNA index(DI),S-period frequency(SPF)and various conditions of Rb invasion to optic nerve(P<0.05) and differentiation of Rb(P<0.01).Conclusion:The study on DNA content may be involved in tumorigenesis and important to the biologic potential and clinical outcome in Rb.
