摘    要:目的 为骨髓造血干细胞 (BMSC)及外周血造血干细胞 (PBSC)的短期保存建立简便有效的方法。方法 采用自体血浆为骨髓单个核细胞 (BMMCs)及外周血单个核细胞 (PBMCs)冷藏营养液 ,直接置BMMCs及PBMCs于 4℃冰箱中保存 ,分别在当天至第 7d选择性测其细胞活力 ,CD+ 3 4 细胞阳性率及粒 -单系造血祖细胞 (CFU -GM )集落形成能力。结果 BMMCs及PBMCs置 4℃冰箱保存 3d后 ,台盼兰拒染率、CD+ 3 4 细胞回收率和CFU -GM回收率分别达 80 %以上 ;保存 4d后 ,上述三项指标仍达 70 %以上。结论 当患者接受高剂量化疗时用此方法能在 4d内安全地保存BMSC或PBSC。

Abstract:Objective To find a simple and effective way of short-term preservation for the bone marrow stem cells (BMSC) and the peripheral blood stem cells(PBSC). Methods The autologus plasma BMMCs and PBMCs were preserved in a 4℃ refrigerator. The viability, CD+ 34 cells and CFU-GM of the BMMCs and the PBMCs were detected at 1~7 days after preservation. Results Three days after preservation at the 4℃ refrigerator, the trypan-blue chromophobic rate, CD+ 34 cells recovery rate and the CFU-GM rate were over 80%, respectively; four days after preservation, the above three parameters still reached over 70%. Conclusion The 4℃ preservation can safely preserve the BMSC or PBSC within 4 days while patients received the high-dose chemotherapy.
