摘    要:目的通过观察外消旋聚乳酸对血管内皮细胞活性的影响,以确定其是否可作为组织工程的细胞支架材料.方法MTT法检测支架材料(PDLLA)浸提液对血管内皮细胞活性的影响,扫描电镜观察血管内皮细胞在支架材料上生长状况.结果在支架对细胞毒性检测中,MTT法显示支架浸提液与正常培养基对细胞活性的影响没有区别,细胞在支架上生长良好,支架材料毒性分级为0~1级.结论PDLLA无细胞毒性作用,可以作为组织工程骨构建的支架

Abstract:Objective To investigate the effects of PDLLA on the bioactivity of vascular endothelial cell in order to confirm whether it could be used as a scaffold.Methods Rabbit vascular endothelial cells (RVEC) were cultured and implanted in PDLLA. The growing conditions of the vascular endothelial cells on the scaffold were observed using scan electron microscope (SEM)and the bioactivity of rabbit vascular endothelial cells cultured with extracts from the scaffold was tested. Results There was no obvious change of cell attachment, morphology, and proliferation after the cells were cultured with extracts or degradation products. The toxicity gradation of the material was 0~1.Conclusion The PDLLA has no cell toxicity, it can be used as an ideal scaffold for tissue engineering.
