大鼠PDLLA/FK506复合神经套管模型脊髓背角c-fos的表达

摘    要:目的:建立大鼠PDLLA/FK506(外消旋聚乳酸/FK506)复合套管坐骨神经再生模型,观察大鼠坐骨神经再生过程中行为学、痛阈以及脊髓背角c-fos表达的改变,研究大鼠PDLLA/FK506复合套管外周神经再生模型中神经病理痛的变化.方法:雄性Wistar大鼠40只,体质量180~200g,随机分成2组,每组20只,A组为坐骨神经损伤PDLLA/FK506套管修复组; B组为假手术组.术后27d起检测行为学、机械刺激阈值、热缩足反射时间及c-fos表达计数.结果:A组术后27d后机械痛阈和热缩足反射潜伏期与B组相差明显,有统计学意义(P〈0.05).A组术侧L4~5脊髓背角与B组相比较,背角浅层c-fos计数增多,有统计学意义(P〈0.05).A组组内c-fos、机械痛阈和热缩足反射潜伏期45d与27d比较均有统计学意义(P〈0.05).结论:大鼠PDLLA/FK506复合套管外周神经再生模型中神经再生过程中诱发脊髓背角浅层c-fos表达,同时出现再生神经支配区域的神经病理痛和疼痛相关行为.
