全反式维甲酸对兔晶状体上皮细胞移行性和黏附性的影响

摘    要:目的 探讨全反式维甲酸(ATRA)对兔晶状体上皮细胞(RLECs)移行性和黏附性的影响及其作用机制,筛选防治后发性白内障的药物。方法 于第二代体外培养RLECs中添加浓度为10-8—10-5mmol/L的ATRA,观察ATRA对RLECs移行和黏附的影响,用SABC法观察ATRA对RLECs细胞网络骨架改变和内源性层粘连蛋白受体(LN-R)的表达。结果 ATRA对RLECs的移行和黏附有抑制作用且其作用呈时间依赖性和剂量依赖性;ATRA可使RLECs骨架重组并使其内源性LN-R表达降低。结论 ATRA能抑制RLECs的黏附和移行;ATRA能调节RLECs的网络骨架分布且明显降低其内源性LN-R的表达;RLECs的黏附和移行与RLECs的LN-R含量分布密切相关。

Abstract:Objective To study the effect of all-trans retinoie acid (ATRA)on the migration and adherence of rabbit lens epithelial (RLECs) in vitro and to evaluate the possiblity of using A TRA for posterior capsular opacification after cataract extraction. Methods RLECs were cultured in vitro and 10~8~10~5 mmol/L ATRA were added to the ealtured cells at the passage two. The effects of ATRA on the migration and adherence of the cells were measured. In addition, the variation of the eytoskeletion of RLECs was assessed by Coomassie BB staining. Endogenous laminin(LN) molecule was examined by SABC immunohistochemistry. Results ATRA could inhibit the motility and adherence of RLECs in dose-dependent and time-dependent manner; ATRA reorganized the eytoaketeton net work of RLECs and decreased the expression of endogenous LN of RLECs. Conclusions ATRA can inhibit the migration and adherence of LECs by modifying the cytoskeleton net work and the expression of endogenous LN of LECs;The quantity and distribution of endogenous LN molecule and their receptors on the LECs may be important factors for promoting posterior capsular opacification by stimulating the motility and adherence of lens epithelial
