IL-6、TNFα对人早孕绒毛滋养层细胞胎盘生长因子表达调控的作用

目的:探讨IL-6、TNFα对原代培养人早孕绒毛滋养层细胞胎盘生长因子(PIGF)表达的调控作用。方法:对胰蛋白酶与胶原酶Ⅰ联合消化、Percoll细胞分离液纯化而得到的滋养层细胞进行原代培养,测定100 ng/mL IL-6、50 ng/mL TNFα对滋养层细胞PIGF表达的影响。结果:酶联合消化及梯度离心能较好地分离早孕绒毛中的滋养层细胞,100 ng/mL IL-6作用于滋养层细胞后PIGF分泌增加,与对照组有显著性差异(P<0.01),TNFα作用于滋养层细胞后PIGF分泌也增加,但与对照组差异不显著(P>0.05)。结论:IL-6对PIGF分泌具有促进作用,TNFα对滋养层细胞PIGF分泌的促进作用不显著,提示VEGF与PIGF可能由母胎界面不同的细胞因子所调控。

Regulatory Effects of IL-6,TNFα on PIGF Expression in Human Primary First-Trimester Trophoblast Cells

Objective To investigate the regulatory effects of IL-6,TNFα on placental growth factor (PIGF) expression in primary first-trimester trophoblast cells. Methods: Cytotrophoblast cells were isolated by trypsin-collagenase digestion and Percoll gradient centrifugation method for primary cell culture. The effects of 100 ng/mL IL-6、50 ng/mL TNFα on PIGF expression were observed. Results: Trophoblast cells could be isolated effectively. PIGF expression increased with 100 ng/mL IL-6, there were significant differences between experiment groups and control groups (P<0.01). TNFα had slight effect on PIGF expression in trophoblast cells, but no difference was found when compared with controls (P>0.05). Conclusion: IL-6, has greater effects on PIGF expression than TNFα, which suggests different regulatory roles of cytokines in PIGF and VEGF expression.
