益气养阴活血方对糖尿病大鼠肝脏病理及肝脏miRNA-126表达水平的影响

摘    要:目的]通过观察益气养阴活血方对大鼠肝脏组织miRNA-126的含量的影响,探讨其改善糖尿病大鼠胰岛素抵抗(IR)的作用机制。方法]12周龄Wistar雄性大鼠60只,随机分组,其中空白组10只,实验组50只。采用高脂饮食加小剂量链尿佐菌素(STZ)诱导形成2型糖尿病大鼠模型,造模成功后,50只糖尿病组大鼠随机分5组,空白组10只、模型组10只予生理盐水,津力达组10只予津力达颗粒、益气养阴活血方大、中、小剂量组各10只,予相应剂量中药灌服,干预4周,常规取血测定血糖、糖化血清蛋白GSP、空腹胰岛素水平等生化指标,取大鼠肝脏切片,HE染色,光镜下观察病理改变;再取一部分肝脏组织,运用实时荧光定量PCR(q RT-PCR)技术,进行miRNA-126含量的测定。结果]益气养阴活血方中、高剂量组和津力达组的大鼠体质量明显降低;益气养阴活血方各剂量和津力达颗粒均可降低大鼠空腹血糖(FBG)、餐后2 h血糖(2 h BG)、GSP;其中益气养阴活血方中剂量效果优于津力达颗粒;益气养阴活血方各剂量组和津力达颗粒均可减轻大鼠肝脏组织病理损害,其中中剂量组效果最为明显。益气养阴活血方各剂量和津力达颗粒均可降低肝脏组织miRNA-126的表达,其中中剂量组效果更明显,优于津力达组。结论]益气养阴活血方能够改善2型糖尿病大鼠血糖代谢紊乱,可改善肝脏的病理损害,其机制可能与降低肝脏中miRNA-126的含量有关。

Abstract:Objective] To discuss the the mechanism of Yiqi Yangyin Huoxue decoction to improve insulin resistance in diabetic rats by measuring the expression level of miRNA-126 using the realtime-quantitative PCR (qRT-PCR) technology.Methods] Sixty wistar male rats of 12 weeks old were randomly divided into two groups, including 10 blank groups and 50 rats in the experimental group.The high-fat diet plus low-dose streptozotocin (STZ) rat model of type 2 diabetes mellitus induced. After the success of the model, 50 diabetic rats were randomly divided into 5 groups, normal group 10 and model group 10 were treated with normal saline, and Jinlida group with Jinlida particles, and TCM low,medium,high dose group, treated with the corresponding dose of Chinese medicine orally, 4 weeks of treatment, blood biochemical indicators of blood glucose, glycosylated serum protein, fasting insulin levels, the liver tissue, HE staining, observation of liver tissue of rats under light microscope. Determination of liver tissue miRNA-126 expression level by RT-PCR method.Results] Jinlida granules and traditional Chinese medicine Yiqi Yangyin Huoxue decoction (TCM) both can significantly reduce the blood sugar,and fasting insulin level of diabetic rats,and TCM medium dose is better than Jinlida granules. Pathological morphology of the Chinese medicine groups of liver cells under light microscope were significantly improved. Both Jinlida granules and TCM can decrease the content of miRNA-126 in liver tissues, with a better effect of the medium dose of TCM.Conclution] Yiqi Yangyin Huoxue recipe can improve the blood glucose metabolism disorder in type 2 diabetic rats, and improve the pathological damage of liver, which may be related to the reduction of the expression level of miRNA-126 in the liver.
