黄丹化瘀饮对大鼠脑缺血再灌注损伤及脑组织BDNF、VEGF表达的影响

摘    要:[目的]探讨黄丹化瘀饮对大鼠脑缺血再灌注损伤后脑组织脑源性神经生长因子(BDNF)、血管内皮生长因子(VEGF)表达的影响。[方法]选取SD(雄性)大鼠60只随机分为5组(每组12只):假手术组、模型组、小剂量组[7.20 g/(kg·d)]、中剂量组[14.40 g/(kg·d)]、大剂量组[28.80 g/(kg·d)]。用线栓法制备脑缺血再灌注模型,在缺血2 h后进行再灌注,再灌注后的24 h将大鼠处死。进行神经行为评分(Zea Longa评分法),用免疫组化法、蛋白质印迹法(Western blot)进行BDNF、VEGF及VEGFR-1蛋白表达检测,观察各组BDNF、VEGF、VEGFR-1表达情况。[结果]模型组脑缺血再灌注24 h后脑梗死体积及神经功能评分与假手术组比较,差异有统计学意义(P0.05);中剂量组、大剂量组脑梗死体积及神经功能评分与模型组比较,差异有统计学意义(P0.05);模型组VEGF、BDNF、VEGFR-1蛋白表达明显高于假手术组,差异有统计学意义(P0.05);中剂量组、大剂量组VEGF、BDNF、VEGFR-1蛋白表达明显高于模型组,差异有统计学意义(P0.05);小剂量组与模型组比较,VEGF、BDNF、VEGFR-1蛋白表达差异无统计学意义(P0.05)。[结论]黄丹化瘀饮用于脑缺血再灌注后脑损伤模型大鼠,能通过上调VEGF、BDNF、VEGFR-1蛋白水平发挥神经保护作用。

Abstract:[Objective] To investigate the effect of Huangdan Huayu decoction on cerebral ischemia reperfusion injury in rat brain derived neural brain tissue growth factor (BDNF), vascular endothelial growth factor (VEGF) expression.[Methods] A total of 60 SD rats (male)were randomly divided into 5 groups (There were 12 rats in each group):the sham operation group, model group, low dose group[7.20 g/(kg·d)], middle dose group[14.40 g/(kg·d)], high dose group[28.80 g/(kg·d)]. A cerebral ischemia reperfusion model was prepared by wire suture method, reperfusion was performed after ischemia 2 h, 24 h rats were sacrificed after reperfusion. Neurobehavioral score (zealonga score) was performed,the expression of BDNF, VEGF and VEGFR-1 protein was detected byimmunohistochemistry and Western blot, the expressions of BDNF, VEGF and VEGFR-1 in each group were observed.[Results] The cerebral infarction volume and neurological function score of model group after cerebral ischemia reperfusion 24 h were compared with those of sham operation group,the difference was statistically significant(P<0.05); the middle dose group, high dose group, cerebral infarction volume and neurological function were compared with the model group, the difference was statistically significant (P<0.05); the expression of VEGF, BDNF and VEGFR-1 in the model group was significantly higher than that in the sham operation group, and the difference was statistically significant (P<0.05); the middle dose group, high dose group VEGF, BDNF, VEGFR-1 protein expression was significantly higher than the model group, the difference was statistically significant (P<0.05); the small dose group was compared with the model group,the expression of VEGF, BDNF, VEGFR-1 protein in the small dose group was not statistically significant (P>0.05).[Conclusion] Huangdan Huayu Decoction reperfusion cerebral injury rats are treated for cerebral ischemia,it can exert neuroprotective effect by up regulating the levels of VEGF, BDNF and VEGFR-1 protein.
