乳酸盆底注射诱导小鼠非细菌性前列腺炎及前列腺上皮内瘤样变

摘    要:目的 乳酸的盆底聚集可能在慢性非细菌性前列腺炎发病过程中起着始动作用,采用盆底注射乳酸诱导小鼠前列腺组织的炎症改变加以验证.方法 对小鼠进行第1、2、4、7天多次盆底乳酸注射诱发前列腺炎症,并于第2、3、5、8天摘取前列腺组织制作组织学标本,进行HE染色观察.结果 组织切片反映出前列腺组织按照盆底乳酸注射时间的延长依次出现了以间质充血水肿,间质及腺泡周围炎性浸润,腺泡腔内出现炎性分泌物为代表特征的炎症病变,最后一组标本中除炎症反应外还观察到了较为明显的腺泡上皮增生,类似于人类的前列腺上皮内瘤(prostatic intra-epithelial neoplasia, PIN)样变.结论 经乳酸盆底注射诱导的小鼠前列腺炎症改变类似于临床患者的病理表现,提示乳酸的盆底聚集在慢性前列腺炎发病机制中起到了重要作用;本实验中观察到的前列腺炎症后PIN样改变为研究慢性前列腺炎与前列腺癌前病变的发生提供了良好的动物模型.

Abstract:Objective Lactic acid is supposed to trigger the process of chronic nonbacterial prostatitis.To prove this hypothesis,lactic acid was introduced as an irritating agent to induce mice's histological nonbacterial prostatitis by pelvic floor injection.Methods Adolescent mice received repeated injections of lactic acid(0.03 mol/L,0.1 ml per time) on the 1st,2nd,4th,7th days.Each of them was sacrificed on the second day after their last injection.All dissected prostates were fixed in formalin and then sectioned for HE staining.Results The prostates undergoing serials of inflammatory changes sequenced as:edema and venous dilation in the stroma,lymphocytes infiltration of the stroma and periglandular region,inflammatory exudates within the acinus.Besides,distinctive epithelial proliferation change was observed on the 8th day which is basically similar to PIN-like lesion in human prostate.Conclusion Lactic acid induced histological prostatitis replicates the pathological changes observed in clinical patients,suggesting the aggregation of lactic acid in the pelvic region plays an important role in the pathogenesis of chronic nonbacterial prostatitis.This study also supports the association between chronic prostatitis and later prostate cancer in a mouse model.
