氧化应激调节泛素羧基末端酯酶L1在人晶状体上皮细胞系SRA01/04细胞株中的表达

摘    要:目的在梯度过氧化氢叔丁醇(tert-butyl hydroperoxide,TBHB)氧化应激情况下,研究泛素羧基末端酯酶L1(ubiquitin car-boxyl-terminal hydrolase L1,UCHL1)和波形蛋白(Vimentin)在人晶状体上皮细胞系SRA01/04细胞株中表达趋势。方法将SRA01/04细胞分为对照组和梯度处理实验组共四组,使用0.10、.5和1 mol.L-1过氧化氢叔丁醇(tert-butyl hydroperoxide,TB-HB)处理人晶状体上皮细胞系SRA01/04细胞03,,6和12 h,采用RT-PCR检测UCHL1和Vimentin在不同时间点的表达情况。结果在0.5 mol.L-1TBHP作用下,UCHL1在SRA01/04细胞中的表达在3 h时降低,6 h时升高,而在12 h时降低;而Vimen-tin在不同浓度TBHP处理的各个时间点表达均无变化(该实验独立重复三次)。结论氧化应激调节UCHL1在人晶状体上皮细胞系SRA01/04细胞株中的表达,提示UCHL1可能在维持细胞内环境稳定和氧化应激中起重要作用。

Abstract:Aim To investigate expression of UCHL1 and Vimentin in human lens epithelial cells SRA01/04 under oxidative stress.Method The expression of UCHL1 and Vimentin was assayed using RT-PCR in human lens epithelial cells SRA01/04 under 0.1,0.5 and 1 mol·L-1 tert-butyl hydroperoxide(TBHB)treatment for 0,3,6 and 12 h.Results Down regulation of UCHL1 expression was detected after oxidative treatment for 3 h and its expression was subsequently restored and increased after oxidative treatment for 6h,then decreased at 12 h after oxidative treatment.But the expression of Vimentin was constant under different concentration of TBHP treatment.Conclusion The expression of UCHL1 was regulated by oxidative stress and it may play an important role in maintaining cell homeostasis under oxidative stress conditions in human lens epithelial cells SRA01/04.
