芎芷地龙汤对偏头痛模型大鼠脑组织痛觉传导通路c-fos和5-HT蛋白表达的影响

摘    要:目的研究芎芷地龙汤对偏头痛模型大鼠的镇痛作用机制。方法将48只健康雄性SD大鼠随机为生理盐水组、模型组、舒马普坦组、中药组,每组12只。皮下注射硝酸甘油制备偏头痛动物模型,经药物干预后在4h、6h取材,测定三叉神经脊束核(SPN5)和中脑导水管周围灰质(PAG)中细胞癌基因fos(c-fos)和5-羟色胺(5-HT)的阳性细胞数。结果 SPN5中蛋白表达:中药组c-fos阳性细胞数减少,4h多于6h,与模型组比较差异有统计学意义(P<0.01);5-HT阳性细胞数增多,4h稍多于6h,与模型组比较有统计学差异(P<0.05或P<0.01)。PAG中蛋白表达:中药组c-fos阳性细胞数减少,4h多于6h,与模型组比较有统计学差异(P<0.01);中药组5-HT阳性细胞数增加更多,6h多于4h,与模型组比较有统计学差异(P<0.01)。结论芎芷地龙汤可以减少由硝酸甘油诱导的c-fos的表达,增加5-HT的表达,进一步减少SPN5神经元的兴奋性,激动5-HT下行镇痛系统,且可持续较长时间,增高痛阈,阻止痛觉信息的传递,起到镇痛作用。
