不同产地麦冬的指纹图谱比较研究

摘    要:目的:建立麦冬两大主产区川麦冬、杭麦冬的正丁醇和乙醚部位的指纹图谱,比较川麦冬、杭麦冬皂苷和黄酮化学成分的异同。方法:分别采用HPLC-ELSD、ESI/TOF-MS方法建立正丁醇部位指纹图谱;以HPLC-UV法建立乙醚部位指纹图谱。结果:在对皂苷部位的检测分析中,HPLC-ELSD、ESI/TOF-MS两种方法分别从不同角度反映了样品的内在信息,川麦冬、杭麦冬的HPLC-ELSD的指纹图谱虽存在共有峰,但谱图整体存在一定的差异;ESI/TOF-MS显示两者在MS1m/z上重叠性较高,在皂苷化学成分“质”上具有较高相似性。HPLC-UV指纹分析表明两者在成分相对集中的谱图区域具有较高相似性。结论:川麦冬、杭麦冬的皂苷和黄酮化学成分具有较高的相似性,HPLC-ELSD、HPLC-UV指纹图谱可用以区别这两个产地的麦冬。

Abstract:AIM:To establish the fingerprints for characterization of the chemical components of maidong(root of Ophiopogon japonicus)in sichuan(Chuanmaidong) and hangzhou(Hangmaidong).METHODS:HPLC-ELSD and ESI/TOF-MS analytical methods were applied to detect n-butanol extracts respectively,and HPLC-UV method was employed to study ether extracts.RESULTS:HPLC-ELSD and ESI/TOF-MS methods provided a wide range of the chemical information of n-butanol extracts.HPLC-ELSD fingerprint patterns,as a whole,were different between Chuanmaidong and Hangmaidong although they shared several mutual peaks.However,ESI/TOF-MS analysis showed close similarity between Chuanmaidong and Hangmaidong in the MS~(1) scan.On the other hand,HPLC-UV fingerprint revealed similarity in the second half of the fingerprint pattern.CONCLUSION:Most saponins and flavonoids contained in the two cultivating areas are similar.Meanwhile,the fingerprint developed by HPLC-ELSD and HPLC-UV methods could differentiate the cultivating areas of the two maidong.
