中药安胎养血合剂对大鼠肾移植急性排斥反应免疫抑制作用的研究

摘    要:目的:研究中药安胎养血合剂(AnTai YangXue Mixture,ATYXM)对大鼠肾移植急性排斥的免疫抑制作用,探讨其与环孢素A(cyclosporine,CsA)是否具有协同作用。方法:建立大鼠异基因肾移植模型,随机分为4组——Ⅰ组,生理盐水对照组(NS),给予NS灌胃,NS 10mL.kg-1.d-1;Ⅱ组,中药ATYXM组,给予ATYXM灌胃,10mL.kg-1.d-1;Ⅲ组,CsA(cyclosprorine,CsA)组,给予CsA灌胃,10mL.kg-1.d-1;Ⅳ组,中西医结合组,给予ATYXM和CsA灌胃,AT-YXM 10mL.kg-1.d-1、CsA2.5mL.kg-1.d-1,两者间隔30min。于术后第5天每组随机取6只处死,取肾脏组织,通过TUNEL法检测凋亡指数,另6只大鼠观察移植肾存活时间。结果:对照组大鼠均在术后5天死亡,平均生存时间为(5.33±0.52)天,ATYXM组(8.67±1.51)天、CsA组(11.17±0.75)天和中西医结合组(13.67±1.63)天的存活时间明显长于对照组(P<0.05)。对照组凋亡指数为(15.90±4.91),ATYXM组为(6.60±2.01),CsA组为(4.00±1.41),中西医结合组为(3.60±1.51),对照组和各个实验组之间比较(P<0.05)。结论:安胎养血合剂通过抑制移植肾细胞凋亡的机制对大鼠肾移植急性排斥反应起到免疫抑制作用,并与环孢菌素有协同效应。

Abstract:Objective:To investigate the immunoinhibitory effects of Chinese Medicine Antai Yangxue Mixture(ATYXM)on acute rejection of kidney transplantation in rats and its synergetic effect with Cyclosporin A(CsA),and explore its immunosuppressive mechanism.Methods: Rat renal transplantation model was established from SD to Wistar rat and randomly divided into four groups,control group,ATYXM group,CsA group,integrated traditional Chinese medicine and western medicine group(combination group).Control group,nomal saline(NS) was administered intragastricly 10mL·kg-1·d-1.ATYXM group 10mL·kg-1·d-1.Cyclosporin A(CsA) group 10mL·kg-1·d-1.Combination group,ATYXM was administered intragastricly 10mL·kg-1·d-1 following CsA 2.5mL·kg-1·d-1.Six recipient rats from each group were sacrificed at fifth day after the operation and the transplanted allografts were studied with histological observation,and apoptotic index(AI) was determined by terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL) method.Results: All recipients in control group died of acute rejection within five days after operation,their survival time was(5.33±0.52) days.That of ATYXM group and combination group were(8.67±1.51) days and(13.67±1.63) days respectively.Apoptotic index of allografted kidney in control group was(15.90±4.91),that of ATYXM group,CsA group and drug combination group were(6.6±2.01),(4.00±1.41) and(3.60±1.51) respectively,there was significant difference between control group and each experimental group(P<0.05).Conclusion: ATYXM's immunosuppressive effect on ecute rejection of kidney transplantation maybe through decreasing cell apoptosis,and combination with CsA had synergistic effects.
