摘    要:目的:平白术多糖分离纯化及单糖组成的研究。方法:水提醇沉得平白术粗多糖(CAMP),三氯乙酸法脱蛋白,双氧水脱色,DEAE-纤维素柱色谱分离纯化得平白术精多糖(RAMP)。红外光谱(IR)、气相色谱(GC)分析RAMP的结构及其组成。结果:DEAE-纤维素柱洗脱分离得三个主要组分,IR显示RAMP具有典型的多糖吸收峰,GC检测得RAMP单糖组成为阿拉伯糖、半乳糖、葡萄糖、甘露糖。结论:GC具有高分离效能、高检测效能的特点,可用于单糖组分分析;IR可用于多糖的分析鉴别。

Isolation, Purification and Analysis of Polysaccharide of Atractylodes Macrocephala Koidz by IR, GC
Abstract:Objective To isolate,purify,and identify the structural features of polysaccharide in Atractylodes macrocephala Koidz.Methods CAMP were prepared by ethanol precipitation of the hot water extract of powdered Atractylodes macrocephala Koidz and deproteinized by TCA method,decolored by Hydrogen Peroxide method.RAMP was separated by DEAE-cellulose.The structural features and the composition of RAMP were determined by IR,GC.Results RAMP were composited of three main ingredients by DEAE-cellulose.RAMP was made up of D-Mannose,D-Glucose,D-Galactose,D-Arabinose.Conclusion GC with high performance of separation and detection can be used for monosaccharide composition analysis.IR can identify polysaccharides.
