IL-18单核苷酸多态性rs1946519和rs360718与宫颈癌的关系

摘    要:目的 探讨IL-18基因单核苷酸多态性rs1946519和rs360718与宫颈癌关系,为研究宫颈癌患病风险奠定基础.方法 常规方法对107例原发性宫颈癌患者和80名正常人群外周血进行基因组DNA抽提,并自行设计引物和探针,用等位基因特异性杂交技术对IL-18基因rs360718和rs1946519位点进行TaqMan探针荧光定量PCR分型,应用x2检验统计分析病例组和对照组等位基因频率、等位基因型频率及其患病风险.结果 宫颈癌患者组与对照组IL-18 rs360718TT和GT基因型频率及其G和T等位基因频率比较无显著性差异(P>0.05);而宫颈癌患者组与对照组IL-18 rs1946519AA、AC和CC基因型频率及A和C等位基因频率差异显著(P<0.05),AC和CC基因型及C等位基因的患病风险分别为2.883,2.750和1.611.宫颈癌与正常人群中IL-18 rs1946519和rs360718位点有6种基因型,比较发现AC TT基因型频率较正常对照组高,有显著性差异(P<0.05).结论 IL-18 rs360718等位基因变异与宫颈癌的易感性无关;IL-18rs1946519与宫颈癌的发生有密切关系,具有AC TT基因型的人发生宫颈癌的危险性相对较高.

Abstract:Objective To investigate the association of interleukin-18(IL-18) rs1946519 and rs360718 single nucleotide polymorphisms(SNPs) with cervical cancer and the risk factors for cervical cancer.Methods The genotypes of IL-18 gene SNPs were detected in 107 cervical cancer patients and 80 healthy female blood donors(control group) by TaqMan and MGB probe assays.The allele and genotype frequencies and risk factors of cervical cancer were analyzed by Chi-square test in both groups.Results No significant differences were found in rs360718 allele and genotype frequencies between patients with cervical cancer and the control group(P>0.05),but the allele and genotype frequencies in rs1946519 as well as the genotypes of rs1946519 and rs360718(AC TT) were found to relate to cervical cancer(P<0.05).Conclusion rs360718 allelic variation in IL-18 gene does not contribute to the susceptibility of cervical cancer in these patients,but rs1946519 allelic variation can be closely related to the pathogenesis of cervical cancer.Women with AC TT genotype are at relatively high risk for cervical cancer.
