摘    要:目的:研究原花青素(PC)对2型糖尿病大鼠坐骨神经传导速度影响。方法:建立2型糖尿病大鼠模型,将所有大鼠随机分为3组:正常对照组(A组)、模型组(B组)、PC治疗组(C组),每组8只。8周末,测定大鼠血清血糖(FBG)、血清胰岛素(FINS)、胰岛素敏感指数(ISI)、丙二醛(MDA)水平及超氧化物歧化酶(SOD)活性,坐骨神经传导速度的变化,常规坐骨神经HE染色。结果:与A组比较,B组的FBG、MDA和FINS水平显著升高,ISI、SOD活性和坐骨神经传导速度明显降低(P<0.01 or P<0.05)。C组的FBG、MDA和FINS水平较模型组降低,而ISI和SOD活性明显增强(P<0.01 or P<0.05)。C组的坐骨神经传导速度快于B组。C组坐骨神经病理学改变较B组轻。结论:PC对2型糖尿病大鼠周围神经病变具有保护作用,该保护作用的机制可能与降低血糖,增强SOD活性,清除过多的氧自由基有关。

Abstract:Objective: To study the effect of proanthocyanidins( PC) on nerve conduction velocity of type 2 diabetes mellitus( T2DM)rats. Methods: Build the model of T2 DM rats. All rats were randomly divided into three groups( n = 8) : normal control group( group A),model group( group B) and PC group( group C). There are 8 rats in each group. After 8 weeks,the fasting blood sugar( FBG),insulin,insulin sensitivity index( ISI),superoxide dismutase( SOD),malonaldehyde( MDA) in Serum blood and conduction velocity of sciatic nerve were detected. HE staining of the sciatic nerve was observed. Results: The level of FBG,FINS,MDA increased significantly while the activity of SOD,ISI and the conduction velocity of sciatic nerve decreased significantly in group B compared with group A( P < 0. 01 or P < 0. 05); group C improved the activity of SOD,ISI and the conduction velocity of sciatic nerve significantly,and the level of FBG,FINS,MDA decreased significantly( P < 0. 01 or P < 0. 05). Pathological change is better compared with group B. Conclusion: Proanthocyanidins could protect peripheral nerve with type 2 diabetes rats. The mechanism of protective effect might be related to the reduction of FBG,enhancement of the activity of SOD,removal of excess oxygen free radicals and reducing the generation of MDA.
