| 摘 要： | 目的制备出一定纯度、免疫反应性好的抗NGAL抗体。方法采用纯的NGAL蛋白免疫动物,制备抗血清,然后进行硫酸胺沉淀法纯化抗NGAL抗体,最后运用SDS-PAGE、Western blotting分别鉴定抗NGAL抗体的纯度与免疫反应性。结果硫酸胺沉淀法纯化抗NGAL,SDS-PAGE检测分析表明抗NGAL抗体的纯度大于70%,Western blotting检测分析表明抗NGAL抗体的免疫反应性良好。结论最后获得了一定纯度、免疫反应性良好的抗NGAL抗体。

| Abstract: | Objective To prepare anti-NGAL antibody with some purity and good immunoreactivity.Methods Animals were immunized with the fure NGAL protein and anti-NGAL serum was prepared. Then anti-NGAL antibody was purified by sedimentation with (NH4)2 SO4. At last purity of the anti-NGAL antibody was analyzed via SDS-PAGE and immunoreactivity of anti-NGAL antibody with NGAL protein was analyzed via Western blotting. Results After purified by sedimentation with (NH4 )2SO4 ,the purity of anti-NGAL antibody was more than 70% via SDS-PAGE and anti-NGAL antibody had good immnoreactivity with the pure NGAL protein by Western blotting. Conclusion Anti-NGAL antibody with definite purity and good immnoreactivity was gained at last. |
