人骨髓间充质干细胞体外培养特性的初步观察

目的　观察人骨髓间充质干细胞 (mesenchymalstemcells ,MSCs)在体外培养时的生物学特性。方法　应用密度梯度离心法将MSCs自骨髓中分离出来 ,利用其贴壁生长的特性加以纯化 ,在常规培养条件下观察其形态学及生长增殖特性。结果　从人骨髓中可分离得到间充质干细胞 ,流式细胞仪检测纯度较高。培养的MSCs呈成纤维细胞样 ,以克隆形式增殖 ,生长曲线示在对数生长期细胞群体倍增时间为 3 3h。结论　骨髓间充质干细胞可有效分离 ,体外培养表现出旺盛的增殖能力。

Characteristics of human bone marrow mesenchymal stem cells in vitro

Objective To investigate some biological characteristics of human bone marrow mesenchymal stem cells cultured in vitro.Methods hMSCs were isolated by density gradient centrifugation and purified on the basis of adherent ability.Morphology and proliferation characteristics were obeserved under routinely incubated conditions.Results Mesenchymal stem cells were isolated from bone marrow.The purify of MSC was high according to flow cytomertic analysis.Cultured MSCs were fibroblast-shaped morphology,proliferated in colonies.During the log phase of growth,cells proliferated with a population doubling time of 33 hours.Conclusion Human bone marrow mesenchymal cells were successfully isolated,and they showed active proliferation capacity in vitro.
