分离测定鱼腥草和山楂果实中槲皮素、芸香苷和绿原酸反向迁移胶束毛细管电泳新方法

摘    要:建立了反向迁移胶束毛细管电泳法同时分离测定鱼腥草和山楂果实中槲皮素、芸香苷和绿原酸的新方法。运行缓冲溶液为70mmol/L十二烷基硫酸钠和30mmol/L NaH2PO4,pH2.1,分离电压-15kV,检测波长214nm。在此最佳条件下,分析物可在15min内实现基线分离。槲皮素、芸香苷和绿原酸的线性范围分别为25-400、25-400、12.5-200μg/mL,相关系数分别为0.9992、0.9992、0.9999,3种分析物的检测限(S/N=3)分别为2.52、3.10、3.64μg/mL。槲皮素、芸香苷、绿原酸的回收率分别为98.5%-103.7%、95.8%~97.6%、99.1%-103.8%。

Abstract:A reverse-flow micellar electrokinetic chromatographic method with direct on-column ultraviolet detection was developed for the separation and determination of quercetin, rutin and chlorogenic acid in herba houttuyniae and hawthorn fruit. The separation conditions of the three analytes were optimized with respect to the concentrations of sodium dodecyl sulfate, NaH2PO4, pH and applied voltage. Baseline separation was obtained for the three analytes within 15 min, using a running buffer containing 70 mmol/L sodium dodecyl sulfate, 30 mmol/L NaH2PO4, pH 2.1 and -15 kV voltage. The linear calibration range was 25-400 μg/mL for quercetin (r=0.999 2) and rutin (r=0.999 2); 12.5-200μg/mL for chlorogenic acid (r=0.999 9). The recoveries were 98.5%-103.7%, 95.8%-97.6% and 99.1%-103.8% for quercetin, rutin and chlorogenic acid, respectively. The detection limits of three analytes (S/N=3) were 2.52, 3.1, 3.64μg/mL, respectively.
