环孢素A对大鼠肝脏缺血再灌注损伤影响的实验研究

摘    要:目的 :探讨环孢素A对肝脏缺血再灌注损伤的作用。方法 :雄性SD大鼠 2 4只 ,实验组 (A组 )术前 4d用环孢素A10mg/kg/d灌胃 ,对照组 (B组 )和假手术组 (C组 )则用生理盐水 5ml/kg/d灌胃 ,建立大鼠部分肝脏缺血再灌注模型。采用生化分析仪 ,肝组织切片HE染色 ,免疫组织化学分别测定血浆谷草转氨酶 (AST)、谷丙转氨酶 (ALT)、乳酸脱氢酶 (LDH)水平 ,肝脏病理改变。血管内皮细胞间粘附分子ICAM - 1的表达。结果 :与对照组比较 ,应用环孢素A的大鼠再灌注 10min后血浆中AST、ALT和LDH的浓度无明显差异 (P >0 .0 5 ) ,再灌注 60min后AST、ALT和LDH的浓度明显低于对照组 (P <0 .0 5 ) ;组织病理损伤减轻 ;肝血窦内皮细胞间粘附分子ICAM - 1的表达减少。结论 :环孢素A可以减轻肝脏缺血再灌注损伤

Abstract:Objective:To investigate the effect of cyclosporine A (CYA) pretreatment on liver ischemia-reper fusion injury.Methods:Twenty-four healthy male Sprague- Dawley rats were divided into three groups: treatment (Group A), control (Group B) and sham (Group C). CYA (10mg/kg/d) was administered orally for 4 days prior to surgery while controls and shams received 0.9% sodium chloride(5ml/kg/d). Rats of Group A and Group B were then exposed to 1h of hepatic ischemia followed by 1h of reperfusion. Group C underwent anesthetization and shamoperation. The serum levels of ALT, AST and LDH were determined in each animals after reperfusion 10min and 60min. At 1 h of reperfusion liver specimens were taken from all animals for histologic analysis, and intercellular adhesion molecule- 1 ( ICAM - 1 ) protein translation level of hepatocyte and endothetium were assessed by immuno histochemistry.Results:Significant decreases in AST, ALT and LDH levels compared to controls and shams in dose CYA rats at 60rain reperfusion(P<0.05).Controls exhibited necrotic hepatocytes and severe inflammatory cell in filtration, while the rats administered CYA demonstrated mild inflammatory cell infiltrate. ICAM- 1 of treatment shepatocyte and endothelim were markedly downregulated. Conclusions:This study indicates that CYA may useful in decreasing initial damage resulting from warm hepaticischemia- reperfusion injury.
