| 摘 要： | 目的在原核系统中表达并纯化GST-Ag85B融合蛋白,制备兔抗Ag85B多克隆抗体.方法首先构建编码结核分枝杆菌Ag85B分泌蛋白的重组表达质粒pGEX-Ag85B,将此质粒转入大肠杆菌,诱导表达融合蛋白并加以纯化;然后将纯化的蛋白免疫家兔,提取家兔的抗血清,纯化制备多克隆抗体并进行鉴定.结果成功构建了原核表达质粒pGEX-Ag85B,转化入大肠杆菌BL21中诱导表达并成功纯化,用纯化的GST-Ag85B融合蛋白成功制备了兔抗Ag85多克隆抗体.结论制备的多克隆抗体为进一步的研究奠定了基础.

| Abstract: | Objective To express and purify the fusion protein GST Ag85B, and to prepare the rabbit anti Ag85B polyclonal antibody. Methods The recombinant prokaryotic plasmid DNA expression vector encoding mycobacterium tuberculosis Ag85B was constructed. Fusion protein GST Ag85B was expressed after IPTG induction, and then isolated and purified from the total proteins of E. coli BL21 transformed by the recombinant plasmid pGEX Ag85B. The polyclonal antibody was obtained by immunizing rabbits with this purified fusion protein, and the quality of the antibody was identified. Results GST Ag85B fusion protein successfully expressed in E. coli and purified polyclonal antibody with high specificity was prepared. Conclusion The rabbit anti Ag85B polyclonal antibody prepared can be used in further studies. |
