氟对鼠成骨细胞BMP表达的影响及锌的拮抗作用

摘    要:目的:研究氟对大鼠成骨细胞中骨形态发生蛋白(BMP-2)表达的影响以及锌制剂能否拮抗氟化物的作用。方法:体外培养SD乳鼠成骨细胞,给予不同剂量氟或/和锌制剂,测定细胞增殖及AKP活性,通过免疫组化方法对各组成骨细胞表达BMP-2进行定量分析。结果:10^-4mol/L ZnSO4及10^-5mol/L NaF能促进细胞增殖,增强AKP活性;10^-3mol/L NaF则抑制细胞增殖及AKP活性;高氟(10^-3mol/L)可促进BMP-2表达,但10^-4mol/L ZnSO4加入高氟组后未发现有抑制BMP-2表达的作用。结论:高氟可促进成骨细胞BMP-2表达,锌制剂能拮抗高氟的毒性作用,但对BMP-2的表达无拮抗作用。

Influence of fluoride on bone morphogenetic protein expression and the antagonistic effect of zinc in rat osteoblasts:an in vitro study
Abstract:Objective To assess the influence of fluoride on the expression of BMPs in rat osteoblast and the antagonistic effect of zinc on the toxicity of fluoride. Methods Osteoblasts obtained from calvarial of sulking SD rats were cultured in medium with different concentrations of NaF and/or ZnSO4. The effects of different concentration NaF and/or ZnSO4 on cell proliferation, alkaline phosphatase(AKP) activity and the expression of bone morphogenetic proteins were determined and evaluated by MTT, enzyme-chemistry and immunohistochemistry measures. Results Both 10-4 mol/L ZnSO4 and 10-5 mol/LNaF promoted cell growth and proliferation and AKP activity in osteoblasts, NaF(10-3 mol/L) could inhibit these effects; but higher dose NaF could significantly trigger the expression of BMP-2 and ZnSO4 added into higher dose NaF could not decrease this effect. Conclusions The results suggest that higher dose NaF might significantly promote the expression of BMP in sucking mice osteoblasts, and ZnSO4 could antagonize the toxicity of high fluoride , but it could not antagonize the increasing expression of BMP resulting from high dose fluoride.
