无忧汤对抑郁模型大鼠海马5-HT1AR和5-HT2AR表达影响的实验研究

摘    要:目的通过对大鼠预见性应激大鼠抑郁模型(CUMS)的复制,观察无忧汤对抑郁模型大鼠海马CA1区5-羟色胺受体(5-HTR)的两个亚型5-HT1AR和5-HT2AR表达的影响。方法随机将60只CL级SD雄性大鼠分为空白对照组,模型组,黛力新组,无忧汤低剂量组、中剂量组、高剂量组6组,每组10只。造模开始前予以适应性饲养1周,造模35 d后,给予相应药物治疗35 d,实验结束后采用HE染色法观察大鼠海马CA1区阳性细胞形态结构和细胞数量的变化;采用免疫组化法,观察无忧汤对大鼠海马CA1区5-HT1AR和5-HT2AR表达的影响。结果经过35 d造模后,药物干预组和模型组大鼠体质量增加量明显低于空白对照组(P<0.05);药物干预组和模型组大鼠的食物消耗量显著低于空白对照组(P<0.05);模型组及药物干预组糖水消耗量明显低于空白对照组(P<0.05);模型组及各给药组大鼠水平得分明显低于空白对照组(P<0.05);模型组及药物干预组大鼠垂直得分明显低于空白对照组(P<0.05)。经过35 d给药治疗后,无忧汤高剂量组与无忧汤低剂量组大鼠相比,体质量增加较明显(P<0.05);无忧汤高剂量组与无忧汤低剂量组大鼠相比,食物消耗量增加显著(P<0.05);无忧汤中、高剂量组与无忧汤低剂量组大鼠相比,糖水消耗显著增加(P<0.05);无忧汤高剂量组大鼠与无忧汤低剂量组大鼠相比较,水平得分增加明显(P<0.05);无忧汤中、高剂量组大鼠与无忧汤低剂量组大鼠相比较,垂直得分显著增加(P<0.05)。与空白对照组相比,模型组大鼠海马5-HT1AR阳性表达率降低(P<0.05),5-HT2AR阳性表达率升高(P<0.05);与模型组相比较,黛力新组和无忧汤中、高剂量组大鼠海马5-HT1AR阳性表达升高(P<0.05),5-HT2AR阳性表达降低(P<0.05)。结论无忧汤抗抑郁机制可能与提高抑郁模型大鼠海马CA1区5-HT1AR并下调5-HT2AR的表达有关。

Experimental study of the effect of wuyou decoction on the expressions of hippocampal 5-HT1AR and 5-HT2AR in depression model rats

Abstract:Objective To observe the effect of wuyou decoction on the expressions of 5-HT1AR and 5-HT2AR in hippocampal CA 1 in the depression model rats through the duplication of chronic unpredictable mild stress(CUMS)in the rats.Methods A total of 60 CL-grade SD male rats were randomized into a blank control group,a model group,deanxit group,wuyou deocoction low-dose group,middle-dose group and high-dose group,10 rats in each one.Before starting modeling,the adaptive feeding was provided for 1 week.After 35 days of modeling,the medication was given for 35 days.At the end of experiment.Using HE staining,the changes of positive cell morphological structure and cell number in hippocampal CA 1 were observed.Using IHC method,the effect of wuyou decoction on the expressions of 5-HT1AR and 5-HT2AR in hippocampal CA 1 were observed.Results After 35 days of modeling,the body weight was obviously lower in the drug intervention groups and the model group as compared with the blank control group(P<0.05).The food consumption in the drug intervention groups and the model group were lower significantly than the blank control group(P<0.05).In the model group and the drug intervention groups,the glucose water consumption was lower significantly than the blank control group(P<0.05).The horizontal score in the model group and each medication group was lower significantly than the blank control group(P<0.05).The vertical score in the model group and each medication group was lower significantly than the blank control group(P<0.05).After 35-day medication,the body weight in the high-dose group was increased significantly as compared with the low-dose group(P<0.05).The food consumption in the high-dose group was increased significantly as compared with the low-dose group(P<0.05).The glucose consumption in the high-dose group was increased significantly as compared with the low-dose group(P<0.05).In the the middle-dose and the high-dose groups of wuyou decoction,the horizontal score was increased significantly as compared with the low-dose group(P<0.05).In the middle-dose and the high-dose groups of wuyou decoction,the vertical score was increased significantly as compared with the low-dose group(P<0.05).Compared with the blank control group,the positive expression of 5-HT1AR was reduced and 5-HT2AR was increased in the model group(P<0.05).Compared with the model group,the positive expression of 5-HT1AR was increased and 5-HT2AR was reduced in the deanxit group and the middle-dose and the high-dose group of wuyou decoction.Conclusion Wuyou decoction displays the anti-depression effect,which is probably related to the up-regulation of 5-HT1AR and the down-regulation of 5-HT2AR in hippocampal CA1 in CUMS model rats.
